Named Manchester Road through St. Louis County, this highway continues west to Linn, Missouri at U.S. Route 50. Much of the road between St. Louis and Gray Summit was the original U.S. Route 66, though this was only temporary until US 66 moved to its later location at what is now Watson Road (Route 366) and Interstate 44. During, and after US 66 was moved off Manchester Road, the road remained part of U.S. Route 50 for many years. Route of the highway

Missouri 100 begins under Interstate 55 at Fourth Street in downtown St. Louis, though there is no connection to the interstate above. It heads west down Chouteau Avenue to Manchester Road where the highway leaves St. Louis and heads into the suburbs. The highway is a relatively narrow, four-lane road for some way through the suburbs. In Kirkwood, at Kirkwood Rd. (known as Lindbergh Boulevard in the other suburbs), MO 100 intersects U.S. Route 61/U.S. Route 67. Two miles further west, in Des Peres, is an interchange with I-270. Because of high traffic, westbound and eastbound lanes are split at this interchange. In Manchester, Route 141 passes overhead with a SPUI for Missouri 100. West, in Ellisville, is the western terminus of Route 340, west of which, the highway becomes an expressway. A few sections of Manchester Road proper split off in this section. At Gray Summit, Missouri 100 passes over Interstate 44 twice, then turns northwest towards the Missouri River. At Washington is an intersection with Route 47, and a few miles west is the northern terminus of Route 185. At Hermann is an intersection with Route 19 and at Chamois is the northern terminus of Route 89. The road then turns southward and ends at Linn. The Missouri River is a tributary of the Mississippi River in the United States. ...


The Lewis and Clark Trail

Most of Route 100 is part of the Lewis and Clark Trail. From Kirkwood Boulevard in Kirkwood to three miles west of Wildwood is one section, where the trail turns off on Missouri Supplemental Route T. Supplemental Route T will eventually end, once again, at Missouri 100 northwest of I-44 and continue down Missouri 100 all the way to five miles north of Linn (where Missouri 100 ends), and the Lewis and Clark Trail turns off onto Supplemental Route C.
