Dornase alfa (Pulmozyme®) is a highly purified solution of recombinant human deoxyribonuclease I (rhDNase), an enzyme which selectively cleaves DNA. Pulmozyme hydrolyzes the DNA in sputum/mucus of CF patients and reduces viscoelasticity in the lungs, promoting improved clearance of secretions. The data suggest that the effects of dornasealfa on respiratory tract infections in older patients (>21 years) may be smaller than in young patients, and that twice daily dosing may be required in the older patients.
Daily administration of dornasealfa for inhalation in conjunction with standard therapies is indicated in the management of cystic fibrosis patients to reduce the frequency of respiratory infections requiring parenteral antibiotics and to improve pulmonary function.
Dornasealfa was tested in a micronucleus (in vivo) assay for its potential to produce chromosome damage in bone marrow cells of mice following a bolus i.v.
